HDAC6-IN-88
HDAC6-IN-88 is a selective HDAC6 inhibitor with an IC50 of 28 nM. HDAC6-IN-88 directly binds to HDAC6 and prevents the deacetylation of α-tubulin (α-tubulin) and Hsp90. HDAC6-IN-88 enhances microglial phagocytosis, promotes microtubule stabilization, and inhibits NF-κB pathway activation. DAC6-IN-88 ameliorates cognitive deficits in Aβ1-42 oligomer-induced mice, reverses HDAC6-related changes in hippocampal biomarkers, and exhibits good biosafety. HDAC6-IN-88 can be used for research related to Alzheimer's disease.

IC50 & Target

HDAC6 28 nM (IC50) |
In Vitro
HDAC6-IN-88 (2 h) directly binds HDAC6 in tissue lysates and protects it from pronase digestion[1].
HDAC6-IN-88 (10 h) prevents cold-induced α-tubulin deacetylation in HT22 neuronal cells[1].
HDAC6-IN-88 (1-100 μM) is well tolerated by HT22 neuronal cells even at high concentrations in the CCK8 viability assay[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
-
Cell Line:HT22 neuronal cells
-
Concentration:1 and 3 μM
-
Incubation Time:2 h pretreatment; Aβ1-42 oligomers 8 h
-
Result:The Aβ1-42 oligomer-induced decrease in acetylated α-tubulin was prevented, increasing the acetyl-α-tubulin/α-tubulin ratio relative to Aβ1-42-treated vehicle cells.
In Vivo
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
-
Animal Model:C57BL/6 N (male, 6-8 weeks, 20-30 g)[1]
-
Dosage:0.5 μg/kg and 1.5 μg/kg
-
Administration:intrahippocampal injection; single co-administration with Aβ1-42 oligomers
-
Result:In the NOR retention phase, Aβ1-42 oligomer-treated mice had a lower discrimination index than controls; a higher discrimination index than Aβ1-42 vehicle was produced.
In the Y-maze, a higher spontaneous alternation percentage than Aβ1-42 vehicle was produced.
Hippocampal acetyl-α-tubulin increased.
Acetyl-Hsp90 was enhanced and p-NF-κB was decreased in hippocampus.
Hippocampal RNA-seq identified 797 DEGs (313 up-regulated, 484 down-regulated); 61 overlapped axon-related genes and 49 overlapped phagocytosis-related genes, with KEGG enrichment in JAK/STAT, NF-κB, Toll-like receptor, and TNF signaling pathways.
