KYS-1 is a specific fluorescent substrate for Cathepsin E, with a K_m of 1.90 μM. KYS-1 is specifically cleaved by Cathepsin E at the Phe-Leu bond under acidic conditions (optimal pH approximately 4.0), relieving fluorescence quenching and releasing a strong signal, thereby enabling measurement of Cathepsin E enzyme activity. KYS-1 can be used for breast cancer research.

Cathepsin E

KYS-1 (200 µM; 10 min) is hydrolyzed by cathepsin E in wild-type mouse serum, where more than 75% of the KYS-1 hydrolytic activity can be immunoprecipitated by anti-rat CatE IgG^[2].
KYS-1 is hydrolyzed by purified recombinant cathepsin E at pH 5.0, and GlcN does not alter this activity^[3].
KYS-1 (6 h) can report CatE enzyme activity in MOG35-55 (HY-P1240)-stimulated mouse neutrophil whole-cell lysates^[4].
KYS-1 (10-40 μM; 10 min) is selectively hydrolyzed by cathepsin E at pH 4.0, with Km values of 1.90, 2.28, and 1.79 μM for human erythrocyte, rat erythrocyte, and rat stomach-derived cathepsin E, respectively, whereas cathepsin D, pepsin A, and cathepsins B, H, and L hydrolyze it to a lesser extent^[1].
KYS-1 (20 μM; 10 min) assay detects cathepsin E in the range of 0.25-4 ng and remains linear under standard conditions^[1].
KYS-1 confirms that cathepsin E activity is completely lost in mammary gland extracts from CatE-/- mice^[7].
