LRRK2-IN-24

LRRK2-IN-24 is an orally active, blood-brain barrier-penetrant and selective LRRK2 inhibitor (IC₅₀ = 1.03 nM). LRRK2-IN-24 inhibits the activity of LRRK2 kinase against wild-type and G2019S mutant forms, and reduces the ratio of phosphorylated LRRK2 to total LRRK2 in mouse hippocampus and cortex. LRRK2-IN-24 can be used in research related to Parkinson's disease.

LRRK2-IN-24 (II-5) is a wild-type LRRK2 inhibitor with an IC₅₀ of 1.03 nM; it is also a G2019S mutant LRRK2 cellular inhibitor with an IC₅₀ of 1.76 nM; it does not inhibit CYP1A2, with an IC₅₀ >30 μM^[1].
LRRK2-IN-24 is metabolically stable in human liver microsomes and mouse liver microsomes, with t_1/2 of > 145 min and 84.0 min, respectively; it exhibits good Caco-2 permeability, with Papp A-B of 7.13 × 10^-6 cm/s and an efflux ratio as low as 1.33^[1].
