Mezigdomide derivative-1 (CC-92480 derivative-1) is a chiral α-methylglutarimide analog of Mezigdomide (HY-129395) and belongs to the CELMoD (CRBN E3 ubiquitin ligase modulator) class. After the introduction of a methyl group to lock the chirality, Mezigdomide derivative-1 almost completely loses CRBN binding and Aiolos protein degradation activity, indicating that this chiral site does not tolerate methyl substitution. Mezigdomide derivative-1 can be used for structure-activity relationship studies.

Mezigdomide derivative-1 (compound 58) exhibits an antiproliferative IC₅₀ of 496 nM in Lenalidomide (HY-A0003)-resistant H929 R10-1 cells, an Aiolos degradation EC₅₀>10 nM, an Aiolos minimum residual protein Y_min=93%, and a CRBN binding IC₅₀>10,000 nM^[1].
