N-De-phenylethyl isohericerin
N-De-phenylethyl isohericerin (NDPIH) is a neurotrophic agent and ERK1/2 activator. N-De-phenylethyl isohericerin activates ERK1/2 signaling in a TrkB-independent manner, and its effects are partially mediated through BDNF/TrkB signaling. N-De-phenylethyl isohericerin promotes axon growth, neurite branching, and growth cone enlargement in central neurons, and enhances memory. N-De-phenylethyl isohericerin can be used for research on neurological diseases.

In Vitro
N-De-phenylethyl isohericerin (1 μg/mL; 24 h) increases growth cone area 3-fold in E18 Sprague Dawley rat hippocampal neurons cultured in serum-containing medium at a concentration of 1 μg/mL, inducing enlarged growth cones with intact cytoskeletal organization[2].
N-De-phenylethyl isohericerin (0.1-10 μg/mL; 24 h) directly induces concentration-dependent neurite outgrowth in low-density, serum-free E18 Sprague Dawley rat hippocampal neurons, with the greatest effect observed at 10 μg/mL[2].
N-De-phenylethyl isohericerin (NDPIH) (10 μg/mL; 24 h) exerts potent neurotrophic activity in primary embryonic rat hippocampal neurons (serum-containing culture), increasing neurite length 3-fold and boosting neurite branching at a concentration of 10 μg/mL for 24 h[3].
N-De-phenylethyl isohericerin (NDPIH) (1 μg/mL; 24 h) increases growth cone size 2- to 5-fold and alters cytoskeletal architecture in primary embryonic rat hippocampal neurons when applied at 1 μg/mL for 24 h[3].
N-De-phenylethyl isohericerin (NDPIH) (0.1-10 μg/mL; 24 h) directly promotes concentration-dependent neurite outgrowth in low-density, serum-free primary embryonic rat hippocampal neurons, with maximal effects observed at 10 μg/mL for 24 h[3].
N-De-phenylethyl isohericerin (NDPIH) (10 μg/mL; 24 h)-induced neurite outgrowth in low-density, serum-free primary embryonic rat hippocampal neurons is only partially mediated by the TrkB pathway, as co-treatment with 0.5 μM ANA-12 for 24 h reduces but does not eliminate the neurotrophic effect of 10 μg/mL NDPIH[3].
N-De-phenylethyl isohericerin (NDPIH) (10 μg/mL; 1 h) activates ERK1/2 signaling independently of TrkB in TrkB-null HEK-293T cells when applied at 10 μg/mL for 1 h[3].
N-De-phenylethyl isohericerin (NDPIH) (10 μg/mL; 1 h) activates ERK1/2 signaling in TrkB and p75 intracellular domain-co-transfected HEK-293T cells when applied at 10 μg/mL for 1 h, with no additional potentiation when combined with 1 ng/mL BDNF[3].
N-De-phenylethyl isohericerin (NDPIH) (0.1-500 μg/mL) does not directly modulate TrkB kinase activity in cell-free assays at concentrations ranging from 0.1 to 500 μg/mL[3].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Line:TrkB-null HEK-293T cells
-
Concentration:10 μg/mL
-
Incubation Time:1 h
-
Result:Increased phosphorylation of ERK1/2 relative to vehicle control, despite absence of endogenous TrkB expression.
-
Cell Line:TrkB-transfected HEK-293T cells
-
Concentration:10 μg/mL (NDPIH); 1 ng/mL (BDNF); 50 μM (ANA-12)
-
Incubation Time:1 h (NDPIH; co-stimulated BDNF); 5 h (ANA-12 pre-treatment)
-
Result:Increased phosphorylation of ERK1/2 to levels higher than those induced by 1 ng/mL BDNF.
Co-treatment with BDNF potentiated ERK1/2 phosphorylation.
Pre-treatment with ANA-12 did not suppress NDPIH-induced ERK1/2 phosphorylation.
Did not alter TrkB phosphorylation levels.

-
Concentration:10 μg/mL (NDPIH); 1 ng/mL (BDNF)
-
Incubation Time:1 h
-
Result:Increased phosphorylation of ERK1/2.
Co-treatment with BDNF did not further potentiate ERK1/2 signaling.
