Ovalitenone
Ovalitenone is a flavonoid found in the plant Millettia peguensis and acts as a CYP1A1/CYP1B1 inhibitor with IC50 values of 0.28 and 0.6 μM, respectively. Ovalitenone significantly decreases the levels of N-cadherin, snail, and slug while increasing E-cadherin, thereby inhibiting the EMT pathway. Ovalitenone also inhibits the regulatory signaling pathways of focal adhesion kinase (FAK), ATP-dependent tyrosine kinase (AKT), mammalian target of rapamycin (mTOR), and cell division cycle 42 (Cdc42). Ovalitenone inhibits the proliferation, migration, invasion, epithelial-mesenchymal transition, and cancer stem cell-like phenotype of lung cancer cells. Ovalitenone can be used in lung cancer-related research.

Cell Line:H460 and A549 cells
-
Concentration:10, 50, 100, 200 µM
-
Incubation Time:24 h (viability); 24, 48, 72 h (proliferation)

Result:Ovalitenone at concentrations lower than 200 µM did not significantly affect viability of H460 or A549 cells at 24 h.
In proliferation assays, ovalitenone at 10, 50, 100, 200 µM had no effect at 24 h; at 48 h, ovalitenone at 50, 100, 200 µM significantly decreased proliferation in a dose-dependent manner; at 72 h, ovalitenone at 10, 50, 100, 200 µM significantly decreased proliferation in both cell lines.

-
Incubation Time:24 h (nuclear staining); 24, 48, 72 h (Annexin V/PI)
-
Result:Ovalitenone at 0-200 µM had no effect on apoptosis or necrosis in H460 and A549 cells at 24, 48, and 72 h by Hoechst 33342/PI staining.
Flow cytometry with Annexin V/PI showed that ovalitenone at 100 and 200 µM did not cause apoptosis or necrosis in either cell line at 24, 48, or 72 h.
