Propyzamide
Based on 1 Customer Validation
Propyzamide (Kerb) is an orally active inhibitor of β-tubulin and AHR signaling pathways. Propyzamide binds β-tubulin at the intradimer interface, disrupting microtubule assembly, leading to mitotic arrest and multipolar spindle formation. Propyzamide enhances C/EBPβ and NF-κB signaling, promoting colitogenic helper T cell differentiation and interleukin-17/interferon gamma-expressing colonic T cells. Propyzamide can be used in research on colitis, inflammatory bowel disease, and liver cancer.

In Vitro
Propyzamide (100 μM; 6 days) inhibits Nicotiana tabacum cv. BY-2 cell division, reaching 53% inhibition in the conductivity assay[6].
Propyzamide (0.75 μM-0.1 M; 24 h) causes complete depolymerization of cortical and spindle microtubules and induces multipolar spindle formation in Nicotiana tabacum cv. BY-2 cells[6].
Propyzamide (3.13-50 μM; 24 h)-induced CYP1A2, CYP2B6, and CYP3A4 expression in differentiated HepaRG cells is suppressed by MC-LA or MC-LR at 0.6 μM, demonstrating functional antagonism[3].
Propyzamide (10 µM) inhibits FICZ-induced AHR activation in HEK293FT/DC2.4 AHR reporter cells[2].
Propyzamide (10 µM) activates VCAM1 and C/EBPβ-dependent pro-inflammatory promoters in reporter assays[2].
Propyzamide (3.13-50 μM; 24 h) alone is not cytotoxic in differentiated HepaRG cells up to 50 μM, whereas combined with 0.6 μM MCs it reduces viability to approximately 80% at 50 μM[3].
Propyzamide (50 μM; 24 h) weakly inhibits PP2A activity in differentiated HepaRG cells, with a relative activity of 0.84[3].
Propyzamide (50 µM; 48 h) disrupts β-tubulin immunolocalization in C. vulgaris spermatids, reducing signals at stages IV-VI and eliminating them from stages VII-X, with nuclear distortion at stages V, VII, and VIII[4].
Propyzamide (50 µM; 48 h) induces extensive ultrastructural abnormalities in C. vulgaris spermatids, including nuclear envelope invagination/fragmentation, disturbed chromatin condensation, dilated ER and autolytic vacuoles, and fragmented microtubular manchette, while flagella remain largely correctly formed[4].
Propyzamide (20 μM; 24-72 h) inhibits Vigna radiata seedling root elongation by approximately 20-25%[5].
Propyzamide (20 μM; 24-72 h) impairs lateral root development and causes root tip thickening in Vigna radiata seedlings[5].

Cell Line:differentiated HepaRG cells
-
Concentration:3.13, 6.25, 12.5, 25, 50 μM (PRO alone); 3.13, 6.25, 12.5, 25, 50 μM (combined with 0.6 μM MCs)
-
Incubation Time:24 h
-
Result:Did not reduce cell viability at the highest tested concentration (50 μM).
Mixtures of PRO (3.13-50 μM) with MCs (0.6 μM) reduced cell viability to approximately 80% at 50 μM PRO + 0.6 μM MCs.
-
Cell Line:differentiated HepaRG cells
-
Concentration:3.13, 6.25, 12.5, 25, 50 μM
-
Incubation Time:24 h
-
Result:Induced expression of all marker genes.
Significant effects on CYP2B6 and CYP3A4 were found from 6.25 μM (BMDL 11.3 and 14.6 μM, respectively).
At the highest tested concentration, expression was increased by 3.4-fold for CYP2B6 and 3.0-fold for CYP3A4.
CYP1A2 and GPX2 appeared to increase in a concentration-dependent manner, but the effects were not significant.
-
Cell Line:differentiated HepaRG cells
-
Concentration:3.13, 6.25, 12.5, 25, 50 μM combined with 0.6 μM MC-LA or 0.6 μM MC-LR
-
Incubation Time:24 h
-
Result:In both mixtures, MCs predominantly drove the observed effects.
Marker gene expression patterns were largely comparable to those of the respective MC alone, opposing those of PRO, resulting in functional antagonism.
Both MCs suppressed the PRO-induced increased expression of CYP2B6, CYP3A4, and CYP1A2.

Animal Model:AB wild-type and lck:gfp (7 days post-fertilization larvae)[2]
-
Dosage:0.2, 1, 5, and 20 μM
-
Administration:in E3 medium; co-administered with 25 μg/mL TNBS; 24-72 h starting at 7 d.p.f.
-
Result:Boosted TNBS-induced intestinal inflammation in zebrafish, as indicated by intestinal scores and expression of pro-inflammatory genes.
