Dabcyl-QALPETGEE-Edans is a synthetic peptide substrate for fluorescence resonance energy transfer (FRET) detection of Sortase A (SrtA) enzymatic activity. Dabcyl-QALPETGEE-Edans is labeled at both ends with the fluorescent group Edans (donor) and the quencher Dabcyl (acceptor), respectively. Dabcyl-QALPETGEE-Edans's central sequence contains a conserved motif specifically recognized by SrtA. Dabcyl-QALPETGEE-Edans demonstrates high sensitivity in vitro assays, enabling precise determination of the half-maximal inhibitory concentration of SrtA .

DiSBAC10 is a voltage-sensitive fluorescent probe used to study cell membrane electrical activity in FRET assays. In a resting polarized cell, DiSBAC10 resides on the outer leaflet of the membrane where it accepts photons from excited fluorescein-labeled proteins and re-emits the photons at a higher wavelength. Depolarization of the cell causes rapid translocation of DiSBAC10 to the inner leaflet of the membrane, thereby increasing the distance between fluorophores and reducing the FRET signal.

PE-Cy5.5 is a far-red emitting tandem dye designed based on the principle of fluorescence resonance energy transfer (FRET), and is widely used in multicolor flow cytometry. PE-Cy5.5 consists of phycoerythrin (PE) as the energy donor and the cyanine dye Cy5.5 as the energy acceptor. Upon excitation by blue or green laser light, PE absorbs energy and transfers it to Cy5.5 via FRET, ultimately resulting in Cy5.5 emitting characteristic far-red fluorescence (Ex/Em = 450-500 nm/698 nm) .

PE-Cy7 is a tandem dye conjugate based on fluorescence resonance energy transfer (FRET). Composed of phycoerythrin (PE) as the donor and Cy7 as the acceptor fluorescent dye, PE-Cy7 is suitable for multicolor flow cytometry, laser scanning cytometry and immunofluorescence applications. PE-Cy7 can be conjugated to antibodies or used in the biotin-streptavidin system to label cell surface antigens in immunophenotyping and multiparametric cell analysis. PE-Cy7 undergoes photon-induced oxidative degradation, whose rate depends on light intensity and storage conditions, leading to reduced FRET efficiency and increased PE leakage. PE-Cy7 can be excited at wavelengths of 488 nm, 532 nm or ~760 nm, and its emission signal can be detected using 780/60 nm, 810/90 nm or 785/50 nm bandpass filters, with a Stokes shift of 300 nm .

T-sCPY-s500R-H is a NADPH-based FRET ratiometric biosensor probe. T-sCPY-s500R-H forms a 1:1 FRET pair upon binding to the eDHFR-HaloTag fusion protein through conjugation of the donor s500R (green fluorescence) and the acceptor sCPY (red fluorescence) with the HaloTag ligand and the eDHFR ligand (TMP), respectively; NADPH binding to eDHFR induces a conformational change that converts sCPY from a colorless spirolactam form to a red fluorescent form, enabling quantification of NADPH levels through the ratio of red/green fluorescence intensity (Ex = 488 or 490 nm; green channel Em = 500-580 nm; red channel Em = 600-700 nm). T-sCPY-s500R-H is applicable to research in fields such as cell metabolism studies, oxidative stress and NADPH homeostasis analysis, tumor metabolism, and NADPH-modulating drug screening .
