Dehydroergosterol is a fluorescent cholesterol analog (Ex/Em = 325/375 nm) used in cholesterol transport, membrane structure research, and live-cell imaging. Unlike cholesterol, Dehydroergosterol contains three additional double bonds, an extra methyl group, and an ergosterol side chain, forming a conjugated system that produces intrinsic fluorescence. In its monomeric form, Dehydroergosterol exhibits excitation peaks at 310-311, 324, and 338-340 nm, as well as emission peaks at 354, 371-375, 390-394, and 414 nm; in its microcrystalline form, Dehydroergosterol shows enhanced excimer emission around 404 and 426 nm. Dehydroergosterol mimics the properties of cholesterol, integrates into membranes and lipoproteins, and localizes to organelles .

Fluorescein-5-thiosemicarbazide is a green fluorescent probe used for carbonyl labeling. The thiosemicarbazide group of FTSC reacts with aldehyde groups at the reducing ends of carbohydrates via reductive amination, and can also undergo chemoselective ligation with ketone groups in systems such as 2-keto Ac4GalNAc to form stable fluorescent derivatives. FTSC is applicable for fluorescent labeling of carbohydrates and enables fluorescence imaging of mucin-type O-linked glycoproteins in the 2-keto Ac4GalNAc metabolic labeling system. Free FTSC exhibits Ex/Em = 495/517 nm in PBS. FTSC is suitable for carbohydrate labeling and cellular glycosylation imaging studies .

Cy3.5 is a cyanine-based red fluorescent dye (Ex/Em ≈ 560-581/600-612 nm). After covalent conjugation with proteins, Cy3.5 exhibits extraordinary fluorescence enhancement with minimal quenching even at high labeling ratios. Cy3.5 can form a FRET dye pair with Cy5.5, which maintains stable FRET efficiency in various biochemical buffers and deoxygenated systems. Cy3.5 is applicable to studies related to fluorescent labeling of antibodies/proteins, single-molecule FRET imaging of double-stranded DNA and nucleosomes, SPION functionalization, in vivo tracing, and other relevant research .

Tetra-sulfo-Cy7 DBCO is a bright and photostable near-IR probe. The Cy7 DBCO is water-soluble, hydrophilic dye often a reagent of choice for assay where minimal non-specific binding and exceptional brightness is required. The fluorescence of Cy7 DBCO is pH insensitive from pH 4 to pH 10 and produces minimal autofluorescence of biological specimens in this region of the spectrum. Fluorescence of this long-wavelength Cyanine dye is not visible to the human eye but is readily detected by most imaging systems.

NtHzBtd is a fluorescent probe for detecting Fe 3+. NtHzBtd is applicable to the selective fluorescent detection of Fe 3+ ions and live cell imaging studies. NtHzBtd can selectively coordinate with Fe 3+ to form a 1:1 complex, triggering chelation enhanced quenching (CHEQ) and intramolecular charge transfer (ICT) processes, which result in fluorescence turn-off, thereby enabling sensitive detection of Fe 3+ and live cell fluorescence imaging. After binding to Fe 3+, NtHzBtd reduces fluorescence intensity, exhibits a rapid response property, with a limit of detection of 0.036 μM and a response time of approximately 55 s. The detection wavelengths are Ex/Em = 334/401 nm (solution system) .

JF629-HTL is a far-red fluorogenic HaloTag ligand. Binding of JF629-HTL to HaloTag protein alters the local environment of the dye, promoting the transition of rhodamine from a non-fluorescent state to a fluorescent state, thereby generating a fluorescence enhancement signal. In the light-controlled psHaloTag system, JF629-HTL shows a 6.7-fold fluorescence enhancement after irradiation at 450 nm. JF629-HTL serves as a research tool for far-red fluorescence imaging and the development of chemogenetic biosensors .

Mito-RhFe is a Fluorescent probe for mitochondrial labile Fe³⁺ monitoring via imaging and flow cytometry. This probe is a rhodamine-based construct with a spirolactam fluorescence signaling group and an N2-hydroxyethyldiethylenetriamine chelator; its delocalized positive charge enables mitochondria-targeting ability in live cells, and it exhibits fine cell membrane permeability. In its native state, it exists in the non-fluorescent spirolactam form, but upon binding to Fe³⁺, it undergoes a ring-opening conversion to the fluorescent rhodamine form, triggering a turn-on fluorescent response; this process is reversible, as the addition of the metal chelator TPEN removes Fe³⁺ and converts the probe back to its non-fluorescent spirolactam form, and re-addition of Fe³⁺ restores fluorescence. The probe shows high selectivity for Fe³⁺ over most other metal cations present in living systems, with a ~90-fold fluorescence enhancement upon binding to 20 equiv of Fe³⁺. Mito-RhFe has excitation/emission wavelengths of Ex/Em = 540/578 nm, with an ~8 nm bathochromic shift in emission upon Fe³⁺ binding, and it can also be excited at 543 nm for confocal imaging with emission detected at 570-620 nm[1].

TPE-2E N-oxide is a fluorescent probe used for hypoxia detection and tumor hypoxia imaging. Due to its active intramolecular motion, TPE-2E N-oxide shows no fluorescence in aqueous solution. In hypoxic environments, its N-oxide group undergoes two-electron reduction by CYP450 reductase and other heme protein reductases expressed under hypoxia, forming hydrophobic aggregates that trigger aggregation-induced emission through the restriction of intramolecular motion. TPE-2E N-oxide exhibits selective lipid droplet localization in cells. The absorption wavelength of TPE-2E N-oxide in dichloromethane is 313 nm; in a 99% hexane/dichloromethane mixed system, its excitation wavelength is 330 nm and emission wavelength is 460 nm. For in vitro cell imaging, excitation is performed at 405 nm, with an emission wavelength range of 430-560 nm .
