Sulfopin (PIN1-3) is a highly selective covalent inhibitor of Pin1 with an apparent Ki of 17 nM. Sulfopin blocks Myc-driven tumors in vivo. The peptidyl-prolyl isomerase, Pin1, is exploited in cancer to activate oncogenes and inactivate tumor suppressors .

ZL-Pin13 is a covalent Pin1 inhibitor. ZL-Pin13 modifies Cys113, blocking Pin1-substrate interactions and peptidyl-prolyl cis/trans isomerase activity. ZL-Pin13 induces partial Pin1 degradation and downregulates MCL-1, cyclin D1, and c-Myc at both the protein and transcriptional levels. ZL-Pin13 can be used in research on triple-negative breast cancer .
