Myrislignan is a PI3K/AKT/NF-κB inhibitor that can cross the blood-brain barrier. Myrislignan exerts anticancer activity by inducing apoptosis and ferroptosis. Myrislignan inhibits the replication and invasion of Toxoplasma gondii; it induces reactive oxygen species (ROS) imbalance, autophagy, and the death of Toxoplasma gondii tachyzoites. Myrislignan inhibits mitochondrial function and ERK1/2 phosphorylation to improve ovariectomy-induced osteoporosis. Myrislignan can be used in studies related to gastric cancer, glioblastoma, osteoporosis, and toxoplasmosis .

Human α-defensin 5 is an orally active antimicrobial peptide with diverse biological activities including antibacterial, antiviral, and antiparasitic effects. Human α-defensin 5 kills UPEC, inhibits bacterial biofilm formation, and disrupts bacterial outer membrane integrity. Human α-defensin 5 is hijacked by Shigella flexneri, enhancing Shigella adhesion, aggregation, and invasion of host epithelial cells, and aggravating histopathological damage in corneal and colonic tissues as well as fever. Human α-defensin 5 inhibits LPS-induced pro-inflammatory cytokine production and NF-κB signaling pathway activation, upregulates the tight junction proteins ZO-1 and occludin, and maintains intestinal epithelial barrier integrity. Human α-defensin 5 inhibits various non-enveloped viruses and exhibits killing activity against Toxoplasma gondii tachyzoites. Human α-defensin 5 reduces UPEC bacterial load in the bladder and urine and alleviates inflammatory damage in a rat catheter-associated urinary tract infection model. Human α-defensin 5 can be used in research related to bacterial infections, viral infections, parasitic infections, and inflammatory bowel disease .

Antiparasitic agent-28 is an orally active, selective inhibitor of Toxoplasma gondii phenylalanine tRNA synthetase (TgcPheRS) with blood-brain barrier penetration. Antiparasitic agent-28 inhibits the growth of T. gondii tachyzoites (TgME49-Fluc strain) an (EC50 = 1 nM) and T. gondii bradyzoites (Tg68nLuc strain) induced by alkaline (EC50 = 3 nM) and glutamine-rich medium (EC50 = 0.1 nM). Antiparasitic agent-28 demonstrates potent anti-toxoplasmosis efficacy in mice infected with TgME49-Fluc tachyzoites. Antiparasitic agent-28 can be used for the study of Toxoplasma gondii infection .

ELQ-121 is a potent inhibitor of the ubiquinol-oxidation (QO)-site of parasites. ELQ-121 has IC50 of 0.05 nM against chloroquine sensitive and multidrug resistant P. falciparum in vitro. ELQ-121 inhibits T. gondii and N. caninum with IC50 below 1 nM in vitro. ELQ-121 suppresses B. besnoiti tachyzoite proliferation with an IC50 of 0.49 nM and induces mitochondrial disruption. ELQ-121 can form polyethylene glycol carbonate ester prodrug which demonstrates in vivo efficacy against P. yoelii in mice. ELQ-121 is suitable for antimalarial research .

CWHM-117 is an orally active aspartic protease inhibitor with selective activity against Toxoplasma gondii. CWHM-117 inhibits the proliferation of Toxoplasma gondii tachyzoites in human fibroblasts, with an EC50 of 2.00 µM. CWHM-117 delays mortality in an acute mouse model of toxoplasmosis. In a chronic mouse model of toxoplasmosis, CWHM-117 reduces brain cyst burden and prolongs survival. CWHM-117 can be used for research on toxoplasmosis .
