Interplay Between Aging and Cancer

Aging is a progressive loss of physiological integrity that impairs function and increases vulnerability to major human diseases, including cancer. Modern aging research organized this process around nine interconnected hallmarks - genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, deregulated nutrient sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, and altered intercellular communication - providing a mechanistic framework for studying age-associated disease. Cancer and aging are now viewed as biologically intertwined rather than simply coincident consequences of time: several hallmarks of aging contribute to carcinogenesis and tumor progression, whereas cancer and its treatment can erode health and accelerate aging. This bidirectional relationship has made aging biology increasingly important for tumor biology, precision oncology, cancer survivorship, and therapeutic decision-making in older populations[1][2].

Cellular senescence illustrates the mechanistic tension at the center of this interplay. Senescence can suppress cancer by imposing essentially permanent proliferation arrest after oncogenic or genotoxic stress, but senescent cells also produce a senescence-associated secretory phenotype (SASP) containing inflammatory and malignancy-associated factors. In premalignant epithelial cells, SASP signaling promoted epithelial-mesenchymal transition and invasiveness through paracrine effects that depended largely on IL-6 and IL-8; oncogenic RAS amplified this phenotype, whereas functional p53 restrained it. Senescent cells persisting within the tumor microenvironment can therefore contribute to tumor progression, metastasis, therapy resistance, and altered immune responses even though senescence initially acts as a barrier to cancer initiation. Aging-related immunosenescence adds another regulatory layer: genomic instability, telomere attrition, epigenetic dysregulation, mitochondrial dysfunction, and chronic inflammation promote immune dysfunction, while SASP, metabolic reprogramming, T-cell exhaustion, myeloid bias, and altered intercellular communication reshape the aged tumor microenvironment and weaken tumor control[3][4][5].

These mechanisms create therapeutic opportunities but also important trade-offs. Many cancer therapies induce senescence in malignant cells as part of their antitumor action, yet they can simultaneously induce senescence in normal tissues. In cancer survivors, treatment-associated cellular senescence is linked with accelerated aging phenotypes and the premature development of chronic disorders, making the biology of aging relevant not only to tumor prevention and treatment but also to long-term survivorship. Senotherapies offer one research direction: senolytic approaches aim to eliminate persistent senescent cells, potentially reducing their effects on the tumor microenvironment and restoring antitumor immune responses. Other emerging approaches described in aging-related cancer research include epigenetic modulation and strategies directed at metabolic and immune dysfunction, although their translation into age-specific oncology requires further validation[4][5][6].

Major gaps remain because the hallmarks of aging form an interconnected network rather than independent pathways, and their relative contributions to aging and cancer remain difficult to separate. Senescence itself is context dependent: the same biological program can arrest premalignant cells yet later support malignant behavior through persistent SASP signaling. Immunotherapy introduces additional complexity because aging can alter immune-cell composition, exhaustion, metabolism, and intercellular communication, while available evidence still supports the need for age-tailored trials and biomarker-driven treatment approaches. Future research therefore needs to resolve when individual aging mechanisms suppress or facilitate cancer, develop reliable biomarkers for biological rather than chronological aging, and determine when senolytic, epigenetic, metabolic, or immune-directed interventions should be combined with conventional cancer therapy. Integrating aging biology with oncology may ultimately improve both tumor control and the management of treatment-associated accelerated aging[1][2][4][5][6].

HY-L034 Anti-Aging Compound Library
Aging is a complex biological process characterized by functional decline of tissues and organs, structural degeneration, and reduced adaptability and resistance, all of which contribute to an increase in morbidity and mortality caused by multiple chronic diseases, such as Alzheimer's disease, cancer, and diabetes. Many theories, which fall into two main categories: programmed and error theories, have been proposed to explain the process of aging, but neither of them appears to be fully satisfactory. The programmed theories imply that aging relies on specific gene regulation, and the error theories emphasize the internal and environmental damages accumulated to living organisms. The damage theories proposed the nine hallmarks that were generally considered to contribute to the aging process: genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, deregulated nutrient-sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, and altered intercellular communication. MCE Anti-Aging Compound Library contains 7,843 compounds, mainly targeting Sirtuin, mTOR, IGF-1R, AMPK, p53, Telomerase, Mitophagy, Mitochondrial Metabolism, COX, Cytochrome P450, Oxidase, etc. This library is a useful tool for anti-aging research.

Particularly within the tumor microenvironment, the abnormal accumulation of lactate not only affects cellular energy metabolism but also promotes disease progression by modulating immune cell functions and mediating protein lactylation, thereby participating in epigenetic regulation and signaling networks. Therefore, systematic investigation of lactate metabolic pathways and their associated metabolites is of great significance for understanding disease mechanisms and developing novel therapeutic strategies. The MCE lactic acid metabolite compound library contains 59 compounds and is constructed around key metabolic pathways involving lactate production, transport, and utilization. This library systematically includes core intermediates from glycolysis, the tricarboxylic acid (TCA) cycle, and the lactate cycle. Focusing on disease-associated metabolic reprogramming, it is suitable for research in oncology, inflammation, and metabolic disorders. The library can be used to elucidate the roles of lactate in tumor microenvironment regulation, immune evasion, and epigenetic modifications (such as protein lactylation). In addition, it provides high-quality small-molecule resources for drug screening, facilitating the discovery of potential modulators targeting key enzymes (such as LDH) or transporters (such as MCTs) involved in lactate metabolism.

HY-L266 PD-1 Compound Library
PD-1/PD-L1 signaling is a central negative regulatory axis that maintains immune homeostasis. It involves the specific binding of PD-1 to its ligands PD-L1/PD-L2 and the subsequent transduction of inhibitory signals, which suppress T cell activation and proliferation, drive T cell exhaustion, and contribute to tumor immune evasion. Dysregulation of this pathway is closely linked to major diseases including cancer, chronic viral infections, autoimmune disorders, and transplant rejection. Immune checkpoint blockade targeting PD-1/PD-L1 has emerged as a leading frontier in cancer therapy and mechanistic research. The MCE PD-1 Compound Library contains 101 structurally defined small molecules, covering modulators related to the PD-1/PD-L1 signaling pathway. It can be used for cancer immunotherapy research, studies of immune resistance mechanisms, and candidate compound screening, providing chemical tools for immune checkpoint mechanism research, drug discovery, and high-throughput screening.

A PD-1/PD-L1 lead-like library was built via a five-step virtual screening process. After collecting 8,947 inhibitors from BindingDB and PubChem and filtering by activity and duplicates, AI similarity screening was performed using GeminiMol. Key pharmacophores were extracted from the PPI interface of co-crystal structures, and molecular was screened via a pharmacophore model, effectively enhancing target activity. Containing 10,000 structurally diverse and drug-like molecules well-matched to the PD-L1 pocket, the library supports virtual docking, high-throughput screening and hit discovery, enabling efficient and rapid development of small-molecule immunotherapies.

HY-P990688 Xaluritamig
Xaluritamig (AMG-509) is a bispecific T cell engager and cytolytic agent with a Kd of 27.6 nM for human CD3ε. Xaluritamig binds to CD3ε via an anti-CD3 single-chain variable fragment (scFv) domain, and to STEAP1 via a bispecific anti-STEAP1 antigen-binding fragment (Fab) domain, thereby recruiting and activating T cells and forming a bridge between T cells and STEAP1-expressing cancer cells. Xaluritamig induces T cell-mediated redirected cytotoxicity, tumor cell lysis, cytokine release, CD8+ T cell activation and expansion, as well as tumor stasis or regression. Xaluritamig contains an Fc domain with no effector function, which prolongs serum half-life, exhibits only minimal activity against cells with low STEAP1 expression and normal cells, and shows extremely low target-related off-tumor toxicity in cynomolgus monkeys. Xaluritamig is used in STEAP1×CD3 XmAb 2+1 immunotherapy and in research on metastatic castration-resistant prostate cancer and Ewing sarcoma.
