Venom-Based Antimicrobial Peptide Discovery

Venom-based antimicrobial peptide discovery treats animal venoms as sources of bioactive peptides that can be isolated, characterized, and optimized as anti-infective leads. Venom peptides evolved for prey capture and defense and act across diverse pharmacological targets, which established venoms as productive starting material for therapeutic discovery. The field builds on decades of venom-peptide pharmacology, but antimicrobial discovery increasingly focuses on antibiotic-resistant pathogens, biofilms, and peptide engineering. Antimicrobial work has progressed from identifying native activities to engineering analogues with improved potency and selectivity. Scorpion studies illustrate this trajectory: bactridines from Tityus discrepans inhibited Gram-positive and Gram-negative bacteria and altered bacterial membrane sodium permeability, while AamAP1 and AamAP2 from Androctonus amoreuxi showed broad antimicrobial activity but also substantial hemolysis. A single His-to-Lys substitution in AamAP1 increased antimicrobial potency while reducing hemolysis at active concentrations.

More broadly, venom-derived peptides are now investigated against antimicrobial-resistant infection and other disease areas, sustaining strong interest in venom peptide libraries as a source of drug candidates. Mechanistically, the clearest recurring theme is direct disruption of microbial membranes rather than inhibition of one conserved protein target. Css54 from Centruroides suffusus adopted an alpha-helical structure in a membrane-mimicking environment and killed zoonotic bacteria by disrupting bacterial membranes; it also inhibited Listeria monocytogenes biofilm formation. Hp1404 similarly showed strong activity against carbapenem-resistant Acinetobacter baumannii with dose-dependent membrane disruption and comparatively low hemolytic activity. Mastoparan-1 from hornet venom rapidly killed methicillin-resistant Staphylococcus aureus, reduced biofilm formation and viability, and did not readily select resistance during repeated sublethal exposure. Wasp peptides Agelaia-MPI and Polybia-MPII also inhibited multidrug-resistant A[1][2][3][4].

baumannii biofilms, with bacterial cell-wall degradation observed, and Agelaia-MPI coating reduced bacterial adhesion on vascular stents. A3, an engineered analogue of the scorpion peptide AamAP1, combined enhanced antimicrobial selectivity with antibiotic synergy against multidrug-resistant Gram-positive bacteria, lowering the peptide MIC to 0.125 micromolar in combination experiments. These examples position membrane permeabilization, biofilm control, and antibiotic potentiation as central application routes for antimicrobial drug discovery and device-associated infection research. The most advanced recent example extends this membrane-centered model into metabolic collapse. AaeAP2a showed activity against carbapenem-resistant A. baumannii, inhibited biofilm formation, disrupted membrane integrity, increased permeability, reduced the NAD+/NADH ratio, dissipated proton motive force, decreased ATP production, and induced reactive oxygen species and hydroxyl radicals; in a mouse peritonitis-associated sepsis model, treatment improved survival and reduced organ bacterial burdens[5][6][7][8][9].

Yet translation remains constrained by the properties repeatedly exposed during discovery: native peptides can be hemolytic or toxic to mammalian cells, and only a limited number of venom-derived peptides have progressed successfully to the clinic. The literature therefore supports future work that couples venomics, proteomics, transcriptomics, and sequence discovery with rational peptide engineering to improve safety while retaining rapid antibacterial action. Comparative testing should prioritize multidrug-resistant pathogens, mature biofilms, compatibility with conventional antibiotics, pharmacokinetic improvement, and in vivo validation. Discovery pipelines also need reproducible links between peptide sequence, structure, membrane selectivity, and biological effect, because antimicrobial potency alone does not establish therapeutic suitability. Because many effective candidates act primarily on membranes, target discovery should distinguish direct lipid-envelope interactions from downstream metabolic effects rather than forcing every venom antimicrobial peptide into a single protein-target model. This distinction can guide safer analogue design, mechanism-based combinations, and more informative preclinical studies across bacterial species and clinically relevant infection models[4][6][7][8][9][10][11].

HY-L067 Antibiotics Library
Antibiotics are types of antimicrobial products used for the treatment and prevention of bacterial infections. Antibiotics can kill or inhibit bacterial growth. Although the target of an antibiotic is bacteria, some antibiotics also attack fungi and protozoans. However, antibiotics rarely have an effect on viruses. The major mechanism underlying antibiotics is the inhibition or regulation of enzymes involved in cell wall biosynthesis, nucleic acid metabolism and repair, protein synthesis, or disruption of membrane structure. Many of these cellular functions targeted by antibiotics are most active in multiplying cells. Since there is often overlap in these functions between prokaryotic bacterial cells and eukaryotic mammalian cells, it is not surprising that some antibiotics have also been found to be useful as anticancer agents. MCE supplies a unique collection of 750 antibiotics, including penicillins, cephalosporins, tetracyclines, macrolides, etc. MCE Antibiotics Library is a useful tool for anti-bacterial or anti-cancer drugs discovery.

HY-L033 Peptidomimetic Library
Peptidomimetics are compounds whose essential elements (pharmacophore) mimic a natural peptide or protein in 3D space and which retain the ability to interact with the biological target and produce the same biological effect. Peptidomimetics are designed to circumvent some of the problems associated with a natural peptide: e.g. stability against proteolysis (duration of activity) and poor bioavailability. Certain other properties, such as receptor selectivity or potency, often can be substantially improved. The design and synthesis of peptidomimetics are most important because of the dominant position peptide and protein-protein interactions play in molecular recognition and signaling, especially in living systems. Hence mimics have great potential in drug discovery. MCE Peptidomimetic Library contains 371 compounds including peptoid, α-helix mimetics, β-turn/sheets mimetics, etc. This library is an indispensable tool of structure-activity relationships in drug discovery.

DILI is thought to occur via several different mechanisms. Among these are direct impairment of the structural (e.g., mitochondrial dysfunction) and functional integrity of the liver; production of a metabolite that alters hepatocellular structure and function; production of a reactive drug metabolite that binds to hepatic proteins to produce new antigenic drug-protein adducts, which are targeted by hosts’ defenses (the hapten hypothesis); and initiation of a systemic hypersensitivity response (i.e., drug allergy) that damages the liver. MCE Drug-induced Liver Injury (DILI) Compound Library contains a unique collection of 642 hepatotoxicity causing compounds and is a powerful tool to research DILI and other drug toxicities. This library can be used to understand the mechanisms of DILI, identify biomarkers for early DILI prediction, and allow timely recognition during drug development, thus finally achieving successful DILI prevention and assessment in the pre-marketing phase.
